twain32.dll should not be in the c:WindowsSysWoW64, it should only be in c:Windows
There was an issue with an older version of the TWAIN 2.0 DSM that could not handle this issue, but it has been resolved.

Make sure you are using the latest version of the TWAIN2 DSM.

Can you post a TWAIN2 DSM log file?
